Drug metabolism in droplet scale with on-chip metabolite identification by ambient mass spectrometry

In this work, we developed and validated a droplet-based drug metabolism screening assay. A digital microfluidic (DMF) approach was adopted to facilitate the actuation of the metabolism reactions in droplet scale. The developed DMF device consists of an integrated low-cost inkjet-printed microheater and a porous polymer based monolith functionalized with immobilized cytochrome P450 (CYP) enzymes. The performance of the droplet-based immobilized enzyme reactors (IMERs) was validated using a fluorescent model reaction detected on-chip using a commercial wellplate reader. For identification of the metabolites, the reaction solution was let dry and the chip was interfaced with mass spectrometry via desorption atmospheric pressure photoionization (DAPPI).
