During the research visit to the Department of Molecular Biology, University of Gdansk, Poland, azulene compounds were studied in biofilm models of Gram-positive bacteria.
The assays used for modeling antibiofilm effects were: measurement of biofilm mass (crystal violet staining determination), cell metabolic activity (resazurin-based assays and use of an ATP-based kit), determination of viable cells by bacterial enumeration on agar plates, life and dead staining (live and dead-Kit based stain) used for quantifications, and also for microscopy studies. Methods to extract extracellular polymeric substances from biofilms were also developed and optimized, followed by methods used for the quantification of the matrix components: carbohydrate, protein, eDNA and lipids.
