This research project includes biochemistry and synthetic organic chemistry. It is aimed at determining the structure of the regulatory domain of protein kinase C (PKC), and developing isozyme specific inhibitors for PKC alpha and PKC delta. The specific objectives of the research are to design and synthesize new PKC inhibitors. One line of experiments will be the development of new inhibitors that are based on our previous templates that provided low-affinity antagonists for PKC alpha. The second line of experiments will take advantage on the new protein structure, once it is available from the X-ray determination, for design and synthesis of new lead molecule that will further be improved in the subsequent rounds of screening and modification. In both types of inhibitor design, we will employ structure-based drug design and solid-phase organic chemistry.
