Porous silicon (PSi) nanoparticles (NPs) are able to penetrate 80% of the cortical depth when injected intraventricularly in the E13 mouse embryo brain. It was possible to track the fluorescently labelled PSi NPs during their propagation in the brain tissue for over 3 days. We further confirmed the particles motion in real-time using in vivo two-photon microscopy. PSi NPs also penetrated the embryonic cortex after injection to the mother. We found PSi NPs in both extra- and intra-cellular compartments. No developmental of macro-morphological changes or increased cell apoptosis was observed after in vivo intracerebroventricular PSi NP administration in mouse embryos. Further investigation of the biomedical potential of PSi NPs for embryonic brain targeted therapeutic approaches would be valuable.
