Clostridium botulinum can thrive under stress conditions, and the capability of C. botulinum strains to grow and produce botulinum neurotoxin under cold stress is a substantial hazard to human health. However, the molecular mechanisms and genetic markers responsible for the cold tolerance of C. botulinum are poorly understood. This chapter focuses on the cold stress response of C. botulinum and the importance of two-component systems in cold tolerance of C. botulinum ATCC 3502 and Beluga E1. The cbo0366–cbo0365 and cbo2306–cbo2307 genes of Group I C. botulinum and the clo3403–clo3404 genes of Group II Beluga E1 show an increased relative mRNA level upon cold shock. Furthermore, an insertional inactivation of genes encoding these systems results in impaired growth in comparison to the wild-type ATCC 3502 or the Beluga E1 strains.
